Top items look like vacuum ports for balancing the carbs. Number one doesn't have one. A vacuum gauge is used to get the readings the same on the other 3 as number one. These should be blocked off.

Hi there

Yes that looks like the choke as indicated.

Yes the second is for coolant to heat the carbs, though I don't know what effect it would have not connecting it?

Are you using the carbs on a bike or car engine, but again can't help, but perhaps someone else can.
